摘要:目的:本研究旨在揭示黄芩苷对氧化应激状态下视网膜色素上皮细胞的保护作用相互作用。方法:AMD模型构建及氧化应激损伤评估;实验组,对照组及黄芩苷干预组分别作用于细胞,检测细胞衰老状态、ROS(reactive oxidative species,ROS)累积、线粒体功能异常、细胞坏死(necrosis)及凋亡(apoptosis)状态,Real-Time定量荧光聚合酶链反应分析SIRT1 mRNA水平。结果:1.氧化应激能造成ARPE损伤,并导致SIRT1 mRNA表达水平下降,2.黄芩苷高、低剂量组能显著改善细胞衰老状态,促进增殖,降低凋亡,并显著改善SIRT1 mRNA水平。结论:ARPE细胞氧化应激损伤中黄芩苷能起到负向调节作用,意味着BC具有抗氧化应激损伤的作用,为临床AMD治疗提供的治疗思路。

Abstract: Objective: The aim of this study was to reveal the protective interaction of baicalin on retinal pigment epithelial cells under oxidative stress. Methods: The oxidative stress injury model of human retinal pigment epithelial cells was established and the damage of ARPE was evaluated by oxidative stress. The experimental group, the control group and the baicalin intervention group respectively act on the cells, detect the state of cell senescence, reactive oxidative species (ROS) accumulation, abnormal mitochondrial function, cell necrosis (NECROSIS) and apoptosis. The mRNA level of SIRT1 was analyzed by real-time quantitative fluorescence polymerase chain reaction. Results: 1. Oxidative stress can cause ARPE damage and decrease SIRT1 mRNA expression level. 2. High-dose and low-dose baicalin groups could significantly improve the senescence state of cells, promote proliferation, reduce apoptosis, and significantly improve the mRNA level of SIRT1. Conclusion: Baicalin can play a negative regulatory role in oxidative stress injury of ARPE cells, suggesting that BC has a strong effect on oxidative stress injury, which provides a therapeutic idea for the clinical treatment of AMD.
