[摘要]
目的: 本研究拟用IL-1β 诱导大鼠脑水肿的动物模型,观察乌司他丁处理后能否改善脑水肿、血脑
屏障通透性,进一步探讨乌司他丁防治脑水肿的机制。方法: Wistar 大鼠随机分为对照组、IL-1β 组、乌司他丁
组,通过脑立体定向仪注入IL-1β,制备脑水肿模型,乌司他丁组在注射IL-1β 之前预先用乌司他丁处理,后分
别于6h,12h,24h,通过干湿重称重法测定各组大鼠的脑组织含水量,通过分光光度法测定各组脑组织中依文思
蓝含量推断血脑屏障破坏情况,通过ELISA 法测定脑组织中炎症因子的含量。结果: IL-1β 诱导后,大鼠脑组织
含水量明显增加,血脑屏障通透性增加,鼠脑组织匀浆上清液中TNF-α、NF-κB 及MMP-9 均明显升高。乌司他
丁治疗后炎症因子明显减少,血脑屏障通透性被破坏得到抑制,脑水肿得以缓解。结论: IL-1β 可以诱导脑水肿
的发生,而乌司他丁具有良好的抑制作用。

[Abstract]
Objective:This study intends to IL-1beta induced animal model of brain edema in rats
and to observe the effect of ulinastatin treatment could improve the cerebral edema and the permeability
of the blood brain barrier and further explore the mechanism of ulinastatin prevention and treatment of
cerebral edema. Methods: Wistar rats were randomly divided into control group,IL-1 beta group and
ulinastatin group ,by injection of IL - 1 beta,preparation of brain edema model. Ulinastatin group
before the injection of IL-1 beta retreated with ulinastatin treatment respectively. At 6h,12h,24h,to
determinate the water content in brain tissue of rats, by spectrophotometric method for the
determination of brain tissue of rats in Evans think blue content to infer the blood brain barrier
damage,the content of inflammatory cytokines were measured by enzyme-linked immunosorbent assay
(ELISA) in brain tissue. Results:IL-1 beta induced rat brain water content increased significantly,
increase the permeability of the blood brain barrier,rat brain homogenate supernatant of TNF- alpha,
NF- kappa B and MMP-9 were significantly increased. Ulinastatin in the treatment of inflammatory cytokines
after significantly reduced the permeability of blood brain barrier damage was suppressed,brain
edema alleviated. Conclusion: IL-1 beta can induce brain edema,and ulinastatin has a good inhibitory
effect.
